The problem occurs because of to many overactive microglial cells caused by an underlying inflammatory state. You definitely want microglia in the brain but a balanced amount. CCL5 causes an increased movement of microglial cells to the brain and increases it's ability to kill neuronal cells. It also increases the release of other inflammatory chemokines from the microglial that attract lymphocytes, antibodies and other cells.

Shut down CCL5 and the other inflammatory chemokines and you stop the flow of excessive microglia and other cells. Those excessive cells in the brain will start dying off. In addition you close off the leaks in the BBB that allow large molecules through.
