I’m honestly not surprised that pro-inflammatory cytokines were upregulated in response to higher doses of Leronlimab.

The multiple and overlapping feedback loops are what maintains homeostasis across multiple biological systems. With CCR5 blockade, the resultant cytokine increase is a response to maintain the signaling, albeit pathologic, which Leronlimab interrupts. An attenuated response to previous cytokine levels may result in increased cyokkne release and upregulation of CCR5 (or parallel/similarly functioning receptors such as CCR2.
I don’t believe anything is as simple with our immune system as one would hope, especially with CCR5 which seems simultaneously more variable and pervasive than other GPCRs.
