Extensive evidence has revealed that MANF is closely associated with several metabolic processes; especially, it has a critical role in modulating glucose homeostasis, insulin sensitivity, lipid metabolism, energy balance, and inflammation. Although the exact mechanisms remain to be explored, the current advances provide a new understanding about the intracellular and extracellular functions of MANF. In addition, although the effect of MANF in different tissues or cell types in metabolic control remain largely undefined, the current data provide encouraging evidence that MANF might be a candidate for treating metabolic disorders, including diabetes, obesity, MAFLD, CVDs, and kidney diseases. Clearly, despite the significant progresses over the past few years, many issues remain unresolved (see Outstanding questions). Extensive studies are warranted to resolve these issues and might provide new perspectives for treating metabolic diseases.
