Good overview.
My understanding or interpretation of 1) The GPS effect.
To me it is two parts, though both I guess classify as the gps effect, the seems slightly different
1) CCR5 makes the tumor more aggressive, the ccr5+ cancer cells cluster together in a different way, which enables the cancer cells to be become metastatic.
Here is an agent based paper explaining what I am trying to say, these projections are ccr5+

2) Once the cancer cells enter the bloodstream and (and thus become) CTCs, being ccr5+ guides them to the most dangerous and deadly possible sites: brain, bones, organs.
