when the stock is down, manipulations must exist.

What I believe is: although we suckers didn't realize it at the time, the move upward (ultimately to nearly 5) on good news was in part manipulated. We thought that the price was finally getting up to the level that the fundamentals of the company warranted, as compared to other small biotechs. However, in reality (looking back at some of the posts from that time) there was some pumping going on by people who later turned to bashing. Those same people have done similar things to other stocks -- for instance, AVXL, which was pumped and dumped at least 3 times by some of the same identities that were on the old CTIX board elsewhere. (Some of them are still at the IPIX board.)
Then, after the stock ran up, various articles and bashers started attacking the stock, and then Mako dropped the bomb with outright lies instead of mere FUD like the earlier ones. The Rosen bogus lawsuit, PR'ed quickly for maximum negative effect, helped make that attack successful.
The continuing campaign of outright lies and FUD by bashers on various social media platforms, some of it by people who even admit they don't own stock, can't be explained easily by anything but a coordinated effort. This is particularly true because some of those identities have accidentally revealed themselves to be one person posting under multiple names. For instance, TheBunny/TheMonolith, scottsmith/phteven/etc. To avoid needless discussion/food fights, I'm not going to post the screen names of some of the others, but one of the smarter ones has one bullish identity and one bearish identity -- and then had the "bullish" one spew a virulent bashing post during the weekend some months ago during which the bashers seemingly were panicking. I'm not sure if he posted that under the wrong identity by mistake (which is how some of the multi-identities reveal themselves, right Bunny?), or if he intended to try to use the weight of his purported bullishness to scare people. In any case, why would people be spending this kind of enormous energy if they were not trying to scare off investors and convince people to sell their stock at low prices? And yes, some of us are spending enormous time trying to counter the lies, because we are invested in a stock that we believe will ultimately have drugs that save people's lives, and in doing so will be the investment of a lifetime.
In any case, when I have watched the trades go by, and looked at the tape of the trades afterward to verify, I have seen many actions which seemed impossible in a stock that was trading without manipulation.
Just one specific example: on one occasion there was a supposed offer of 70,000 shares. One of our long-time investors put an order in for a pretty good number of shares at the ask. The 70k "ask" vanished before filling him. I was watching my broker's so-called Level 2 at the time and I saw the 70k ask vanish from the screen. I've seen "asks" disappear when orders came in, and others have told me they have also tried to buy at the ask and not gotten filled as the ask disappeared.
Is that normal stock behavior?
I have personally sat and waited for minutes (and I don't mean it just seeming like a long time, I mean I watched the clock) for my order AT THE ASK to get filled. If it's a legitimate ask, it should get filled within a second or so, right? But no, I have waited several minutes on numerous occasions when trying to buy at the ask. Others have PM'ed me that they've had the same experience.
I've also seen my order sit there unfilled, while some order was filled at a lower price.
Then there are those end-of-day trades when people told me they wanted to be sure they got a fill, so they put the order in either at the ask or at market.
They got filled UNDER the ask.
So, that's why it seems that there is some manipulation going on.
