TKMR Yup, blowing past $30.00 a share this week....
Most likely the price will gap up on Monday due to a very strong technical breakout with very high volume last Friday. Also, the strong buying in the last 10 minutes of trading. Friday was day 2 of 3 of a textbook technical breakout.

Both traders and investors, plus shorters who want in (or cover) will put in market orders prior to the opening. This will drive the opening price higher. Watch the Level 2 on the pre-market as the bids/asks will jump around.
I don't have any yet, but I want to buy a boat load for cheap. Do you think that's possible.

I hope tomorrow there is a pullback in the morning.
Actually, it doesn't take them that long. It only takes the competitor 2 weeks to create another lot. However, I believe it will take two months to get a couple hundred doses.

However, as a patient, I'd prefer to take TKM-ebola since it has a better success rate in the pre clinicals. Substantially better.

I'm not sure why the two missionaries chose to use the competitors therapy other than they wanted to stay with a company in the US since they are from the US? But, it wasn't the logical choice based on pre clinical results.

The FDA has now given the ok for TKMR to treat diseased patients with TKM-ebola. We now wait to see what WHO and the international community decides is the best route. My bet is that multiple companies end up treating sick ebola patients. But, TKMR is set up to get in there first. They are the closest to being ready since they are the only company to have already started a phase I safety trial in healthy humans.

The trial was put on hold until some questions could be resolved, but, that was changed to a partial hold allowing the treatment of diseased patients.

There may in fact be a dip if many misunderstand and actually think the competitor has a cure. But, that won't last long as reality sets in.

We don't know if TKM-ebola works as well in humans as it did in primates, but, we do know it works better in the primates than the competitors therapy did!

Comparing ZMapp and TKM-ebola

Looking at the virus targets for ZMapp compared to TKM-Ebola, TKM-Ebola has a much stronger approach to fighting the virus:

*** ZMapp is a selection of the 3 best components of mb-003 and ZMab ZMapp info (pdf)
-- Mapp Biopharma's MB-003 is a combo of 3 MAbs - Olinger 2012 abs:13C6, 13F6, and 6D8) targeting "3 distinct, non-overlapping EBOV glycoprotein (GP) epitopes. Only 13C6 binds to secreted GP (sGP), which has been speculated to act as a decoy for protective antibodies. 13C6 and 6D8 neutralize virus in the presence of complement, and 13F6 has no neutralizing activity."
-- Defyrus' ZMAb is a combo of 3 MAbs - Qiu 2012 abs : 1H3, 2G4, and 4G7 targeting EBOV GP.

ZMapp being a selection of components from MB-003 and ZMAb, its 3 MAbs all target GP.
So, ZMapp simply impairs virus budding from infected cells and makes the virus more visible (GP helps evade immune response by epitope masking and steric shielding).

*** TKM-Ebola is a combo of 3 siRNAs formulated in SNALPs - Geisbert 2010 abs : targeting ZEBOV L polymerase (EK-1 mod), viral protein (VP) 24 (VP24-1160 mod), and VP35 (VP35-855 mod).
So, TKM-Ebola stops virus reproduction (polymerase) and enhances the immune system response (vp24 and vp35 impair adaptive immune response by diminishing IFN response in several ways).
Additionally, LNP formulation is uptaken by key cells involved in the infection (Tekmira presentation May 2014, slide 16).

So, for all the buzz currently generated around ZMapp, I am not surprised about the so-so past results in NHP and have some doubts about what happened in humans.
With its heat-friendly lyo presentation and proven in NHP efficacy, TKM-Ebola should stay on the forefront of the Ebola fight.
For the long term, in view of HIV, which is know using quadruple combos, I would see TKM-Ebola as a key component in future Ebola combo treatments.


On another note: It is interesting to see suddenly a bunch of FUD messages. Sounds like the 5.3 M shorts from Friday, according to ShortAnalytics, are frightened...

The W.H.O has only talked about a GSK drug set to start clinical trials in September and to be ready by July to end of next year. Zmapp is not feasible for large scale production on short notice for this type of outbreak.
