Scientists from Palacký University Olomouc and the Technical University of Ostrava have pioneered a groundbreaking method to create a new generation of antibiotics. Radek Zbo?il, a physical chemist involved in the research, explained, "The novel material we created effectively kills and prevents the growth of all bacteria examined, including those with high resistance. It functions at low concentrations that are harmless to human cells. Importantly, bacteria cannot develop resistance against this antibiotic, addressing one of the most critical challenges faced by modern medicine. These exciting findings indicate that atomic antibiotics could soon be implemented in practical applications."
To develop this antibiotic, scientists applied their deep knowledge in the fields of graphene chemistry and atomic engineering. They employed a graphene derivative enriched with oxygen and nitrogen atoms to chemically introduce manganese. This transition metal plays various roles in metabolism, bone formation, and the regulation of blood sugar, thereby aiding cellular protection against oxidative stress.

"Our strategy was to disrupt one of bacteria's strongest defenses—the carbohydrates present in their cell walls and membranes, which are critical for survival. These carbohydrates provide protection, facilitate substance transport, enable environmental communication, and serve as energy reserves. By chemically linking manganese to specific carbohydrate configurations, we impeded these essential functions, ultimately causing cell death. The unique graphene carrier ensures that manganese ions are effectively delivered to the bacterial surface, enabling a targeted assault on carbohydrate molecules," Zbo?il elaborated.

The new material demonstrates exceptional effectiveness against bacteria that pose challenges for existing antibiotics. David Paná?ek, the first author of the study, noted, "We recorded superb bactericidal effects against all bacteria from the ESCAPE group, which includes particularly resistant bacterial pathogens known to complicate treatment and increase infection risks, especially in hospital environments. This atomic antibiotic uniquely managed to eliminate all resistant strains in our testing, outperforming conventional antibiotics."
In-Vivo Testing and Future Applications
The researchers did not stop at lab tests. Collaborating with their international partners, they tested the atomic antibiotic in mouse models. "In in-vivo tests, skin infections caused by resistant strains of Staphylococcus aureus were treated swiftly and effectively, with a significant reduction in inflammation markers. We are contemplating further applications for wound dressings or antibacterial coatings on artificial materials, highlighting the immense potential to inhibit bacterial biofilms on devices like artificial joints or stents," stated Milan Kolá?, a microbiologist and Dean of the Faculty of Medicine and Dentistry at Palacký University Olomouc, who was integral to the study.
