"CCR5 is expressed in melanoma. The cold tumor problem exists there too. A meaningful portion of melanoma patients don't respond to checkpoint inhibitors even though Keytruda is already an approved standard of care in that indication. If leronlimab's Prime and Pair mechanism converts cold tumors to hot on demand, melanoma is exactly the kind of indication where that capability would matter most.

And the market context is hard to ignore. The U.S. spends $8.9 billion annually treating skin cancer, with melanoma driving a significant share of that burden. ICIs already account for 44% of melanoma spending among Medicare patients. The reimbursement infrastructure is already there. The annual cost of treating newly diagnosed melanoma cases alone is projected to reach $1.6 billion by 2030.

None of this is to get ahead of where the data actually is. ESMO in October is the moment that matters. The confirmed ORR needs to come in. Everything has to replicate at scale. I'm not suggesting melanoma is next on the timeline. I'm asking whether the biology points there if the mechanism continues to hold.

Dr. Lalezari mentioned two undisclosed solid tumor ITTs in development with updates expected this summer. Whether melanoma is one of them I don't know. But it fits the profile almost perfectly."
