"The efficacy of checkpoint blockade immunotherapy targeting PD-1/PD-L1 interactions is enriched in patients having a T cell-inflamed tumor microenvironment (TME). As such, understanding the biology of the process of tumor cell-immune system interactions is paramount, and fundamental principles are guiding the development of novel strategies to modulate the TME to catalyze the transition of “cold” tumors into “hot”. Non-T cell-inflamed tumors lack essential immune components for anti-tumor immunity, including key subsets of dendritic cells, innate immune activation including type I IFN responses, and chemokines for T cell recruitment. Thus, multiple approaches are being pursued to bring these factors into the cold tumor TME. However, it also has become clear that non-T cell-inflamed tumors are enriched for various subsets of suppressive macrophages that impede anti-tumor immunity in a dominant-negative fashion, and novel targets and strategies are being uncovered to shift myeloid cell phenotypes to be immune-supportive. This session will discuss biologic features of tumor/host interactions, including recent data on macrophage modulation".
