Could the BBB issue with Leronlimab and Mice simply have something to do with the fact that Leronlimab will stop the inflammation very quickly and therefore the leaking of Leronlimab would dissipate relatively early in the treatment.
Back when leronlimab was first discussed years ago I thought that was a possibility. It would be a seesaw effect with leronlimab passing through at a high rate reducing inflammation and lowering BBB leakage then an increase in inflammation and leronlimab rolls through unimpeded. You would see some leronlimab getting through no matter what due to vascular injury. It would certainly explain the 70% rather than 100% occupancy rate. Each cycle could lower inflammation further. One thing to note is that BBB leakage itself adds to the inflammation in the brain so reduced leakage is beneficial.
