With dosing every 7 days, with a 10 day half-life, and the multiplier the sum of all the dosing, i.e. current multiple of dose in the system. So day 7 on the second dose of 525mg, you would have 1.62x525mg in the system, the 525mg dose at day 7 and .62 of 525mg dose from day 0, (1/2)^(7 day /10 day half-life)=.62.
Anyway with a 4 week overlap at 525mg (day 28), that would be 1244mg in the system, so 1244/700 = 1.77 (so around 700mg at 2 weeks dose day 0, 7, 14).
The pivotal trial ind they submitted did something similar, 700mg first 10 weeks, then switch to 525mg. I didn't like that, because the steady state dose would drop by 25% with the drop from 700 to 525, pushing some the failures that would have happened on 525mg but not at 700mg to fail at week 10 instead of earlier. The steady state dose ~ 2.57x525mg = 1349/700=1.92, so they could go with 700 to day 14 and switch to 525mg + drop haart, i.e. 2 week overlap on 700mg
