MediciNova Announces Positive Interim Safety and Clinical Outcomes Data From Clinical Trial of MN-166 (ibudilast) in ALS Presented at the 26th International Symposium on ALS/MND in Orlando, FL

LA JOLLA, Calif., Dec. 13, 2015 (GLOBE NEWSWIRE) -- MediciNova, Inc., a biopharmaceutical company traded on the NASDAQ Global Market (NASDAQ: MNOV ) and the JASDAQ Market of the Tokyo Stock Exchange (Code Number:4875), announced that principal investigator Dr. Benjamin Rix Brooks, Director, Carolinas HealthCare System's Neuromuscular/ALS-MDA Center, today presented interim data from MediciNova's ongoing clinical trial of MN-166 (ibudilast) in amyotrophic lateral sclerosis (ALS) at the 26th International Symposium on ALS/MND (amyotrophic lateral sclerosis/motor neurone disease) in Orlando, FL.  The interim analysis includes a total of 36 subjects, and 33 of those subjects completed 6 months of treatment on study drug (MN-166 or placebo in a 2:1 ratio).

Major highlights from the presentation, “Adaptive Design Single Center Phosphodiesterase Type 4 (PDE4) Inhibitor – Ibudilast (MN-166) Phase 1b/2a Clinical Trial [NCT02238626] for Amyotrophic Lateral Sclerosis (ALS) Patients [1] Not Requiring Non-Invasive Ventilation (no-NIV) up to 5 years and [2] Requiring Non-Invasive Ventilation (NIV) up to 10 years from Disease Onset,” include the following:

  • The rate of decline in the Amyotrophic Lateral Sclerosis Functional Rating Scale-Revised (ALSFRS-R) score, a measure of functional impairment, from Baseline to Month 6 for the MN-166 and placebo subjects combined is below rates observed historically in other ALS studies.
  • Slow vital capacity (SVC), a measure of respiratory function, declined by 1.50% predicted/month for the no-NIV cohort (MN-166 and placebo subjects combined) from Baseline to Month 6, which is below rates observed historically in other ALS studies.
  • Non-invasive ventilation (NIV) utilization was lower in clinical trial participants when compared with the deployment of non-invasive ventilation in a sample of patients referred to ALS Multidisciplinary Clinic with vital capacity greater than 60% predicted (an inclusion criterion for this study), and the difference came close to reaching statistical significance (p=0.07).
  • No cluster of adverse events was differentially present in MN-166 treatment and placebo treatment subjects.
  • Safety review concludes that the clinical trial may continue as planned.  MN-166 treatment will be given to subjects in the open-label extension for an additional 6 months.

About the ALS Trial

MediciNova, in collaboration with Dr. Benjamin Rix Brooks, Director, Carolinas Healthcare System’s Neuromuscular/ALS-MDA Center, is currently evaluating MN-166 (ibudilast) in both early and advanced stage ALS patients.  This ongoing trial is a randomized, double-blind, placebo-controlled study which includes a six-month treatment period followed by a six-month open-label extension. The study is evaluating several efficacy endpoints including functional activity (ALSFRS-R), respiratory function and muscle strength in subjects with ALS. The study plans to enroll 60 ALS patients without NIV (Non-Invasive Ventilator) and an additional 60 ALS patients with NIV.

About ALS

Amyotrophic lateral sclerosis (ALS), also known as Lou Gehrig's disease, is a progressive neurodegenerative disease that affects nerve cells in the brain and the spinal cord. The nerves lose the ability to trigger specific muscles, which causes the muscles to become weak. As a result, ALS affects voluntary movement and patients in the later stages of the disease may become totally paralyzed. Life expectancy of an ALS patient is usually 2-5 years. According to the ALS Association, there are approximately 30,000 ALS patients in the U.S. and approximately 5,600 people in the U.S. are diagnosed with ALS each year. Riluzole is the only pharmaceutical treatment approved for ALS, but it has limited efficacy.

MN-166 (ibudilast) has been marketed in Japan and Korea since 1989 to treat post-stroke complications and bronchial asthma. MediciNova licensed MN-166 (ibudilast) from Kyorin Pharmaceutical for potential utility in relapse-remitting multiple sclerosis (RRMS). Intellectual property was additionally established or obtained by MediciNova in progressive MS and other neurological conditions.
