NuCana Highlights Promising Results from Recent Cancer Study

NuCana Showcases Promising Data from Cancer Treatment Trials

Recently, NuCana plc (NASDAQ: NCNA) revealed significant initial findings from its ongoing Phase 1b/2 modular study focused on NUC-3373, a treatment aimed at patients with advanced solid tumors. This study explores the effects of NUC-3373 when combined with two different therapies: pembrolizumab and docetaxel. The data, which has been published on MedRxiv, highlights noteworthy outcomes for patients who have exhausted other treatment options.

Significant Tumor Volume Reductions Achieved

The primary aim of this study, known as NuTide:303, is to measure the efficacy of NUC-3373 in combination with an anti-PD-1 antibody, pembrolizumab, as well as docetaxel. In the initial Module 1, researchers focused on 12 patients suffering from various solid tumors. Most of these individuals had previously undergone PD-(L)1 based therapy without success.

Pivotal Patient Outcomes

Results displayed encouraging signals of anti-cancer activity, showcasing that 2 patients experienced confirmed Partial Responses while an additional 4 achieved Stable Disease. This led to an impressive overall objective response rate of 22%, with a disease control rate of 67% among those evaluated for efficacy.

Case Study Highlight: NUC-3373 plus Pembrolizumab

Among the patients in this module was a 72-year-old individual diagnosed with urothelial bladder cancer linked to Lynch syndrome. This patient had previously gone through several treatments, including gemcitabine and atezolizumab. Remarkably, following NUC-3373 plus pembrolizumab therapy, they achieved a 100% reduction in their target lesions and continue their treatment more than 10 months later.

Module 1 Continued Success

Another noteworthy case involved a 75-year-old patient battling metastatic cutaneous melanoma. Previously treated with pembrolizumab and dabrafenib, this individual achieved an 81% reduction in the target lesion following the combination therapy and remains stable after over a year.

Exploring Lung Cancer Treatments

In Module 2, the focus shifted to patients with advanced lung cancer. This segment included 4 participants with non-small cell lung cancer (NSCLC) or pleural mesothelioma who either faced disease progression or were intolerant to prior chemotherapy regimens.

The Challenge with Docetaxel

Docetaxel is known as the standard care for NSCLC, yet its benefits are often limited, yielding modest progression-free survival durations of only about 3-4 months coupled with significant side effects. After experiencing toxicity challenges with docetaxel, enrollment was paused, but promisingly, 2 patients still showed prolonged Stable Disease.

Case Study Highlight: Treatment with Docetaxel

One significant case included a 60-year-old patient with pleural mesothelioma who endured multiple previous treatments. After engaging in the NUC-3373 plus docetaxel regimen, they managed to maintain Stable Disease for over 13 months, a notable feat in this challenging oncology environment.

Future of NUC-3373

Professor David Harrison, NuCana’s Head of Translational Medicine, expressed optimism regarding the results, stating that these findings further validate the anti-cancer potential of NUC-3373. This innovative approach, combining NUC-3373 with established treatments, is paving the way for enhanced therapeutic strategies aimed at resistant cancer cases.

Insights From NuCana

NuCana is deeply committed to improving treatment outcomes in oncology through the application of its proprietary ProTide technology, transforming conventional chemotherapy agents into more effective treatment options. With an advanced pipeline featuring both NUC-3373 and NUC-7738, the company continues to focus on developing solutions that effectively target cancer at its core.

Looking Ahead

With ongoing studies and data collection, NuCana is set on a path to reveal more insights and strategies designed to enhance the effectiveness of cancer treatment targeting particularly resilient tumor types. The significance of their findings is evident not only in the promise they hold for current patients but also their potential implications for future cancer therapies.
