NuCana Provides Update on Phase 2 Randomized Colorectal Cancer Study
NuTide:323 Study Discontinued Following Initial Analysis
Ongoing Studies Show Promise with NUC-7738 and Pembrolizumab
NuCana plc (NASDAQ: NCNA) has shared important updates about its clinical trials focused on innovative treatments for colorectal cancer. The company has made the decision to discontinue the NuTide:323 study based on recommendations from the Steering Committee after a planned initial analysis. Although the control arm showed favorable conditions, it was concluded that the combination treatment of NUC-3373 with leucovorin, irinotecan, and bevacizumab (NUFIRI+bev) was unlikely to achieve the primary goal of superior Progression Free Survival (PFS) compared to the control regimen, which consists of 5-FU, leucovorin, irinotecan, and bevacizumab (FOLFIRI+bev). Throughout the trial, both treatment regimens were generally well tolerated, with only a small percentage of patients opting to discontinue due to side effects.
Hugh S. Griffith, the Founder and Chief Executive Officer of NuCana, expressed his disappointment regarding the findings of the NuTide:323 study. He acknowledged the difficulties involved in developing effective treatments for complex cancers like metastatic colorectal cancer but reassured stakeholders that valuable insights were gained from this study. Griffith emphasized the company’s commitment to further exploring potential treatment options using the data collected.
Professor Josep Tabernero, MD, PhD, who heads the Medical Oncology Department at Vall d´Hebron University Hospital and chairs the Steering Committee for the NuTide:323 study, shared their goal of establishing NUC-3373 as an effective alternative to traditional therapies such as 5-FU. He reflected on the disappointment felt by the study team regarding the results, which did not meet their initial expectations.
Despite the challenges faced with the NuTide:323 study, Griffith assured that this development will not impact the ongoing NuTide:303 study. In this trial, NUC-3373 is being tested in combination with pembrolizumab for patients with solid tumors, as well as in another cohort with docetaxel for lung cancer patients. Additionally, there is growing excitement around NUC-7738, a novel anti-cancer agent that effectively modifies gene expression within cancer cells while targeting various aspects of the tumor microenvironment. Upcoming presentations at the ESMO annual conference will provide detailed information from the Phase 2 segment of the NuTide:701 study, focusing on NUC-7738 combined with pembrolizumab for melanoma patients.
About NuCana
NuCana is leading the way in biopharmaceutical innovation, aiming to improve cancer treatment outcomes through its proprietary ProTide technology. This transformative approach is designed to enhance the delivery and effectiveness of established chemotherapy agents, particularly nucleoside analogs, which often struggle with efficacy and safety. NuCana's diverse pipeline includes treatments like NUC-3373 and NUC-7738, with NUC-3373 currently being evaluated in two clinical studies: the NuTide:302 study, which focuses on metastatic colorectal cancer, and the NuTide:303 study for advanced solid tumors.
NUC-7738 represents a significant advancement in cancer therapy, targeting RNA polyadenylation and greatly influencing gene expression while addressing the tumor microenvironment. The ongoing Phase 2 trials for NUC-7738 aim to establish its efficacy as both a standalone treatment and in combination with pembrolizumab for patients with various advanced solid tumors.
