There are several questions with the vaccines:
(1) how long does immunity last?
(2) what about mutations - will it be effective against those?
(3) can give booster doses or booster with another mutant/variant?
(4) long term safety?
#s 1 and 4 no answer until later. #2 they studied in vitro with mostly good results (except as you noted 6x less effective neutralization from elicited antibodies from one spike protein variant to some new variants) but will that predict real world efficacy?
MRNA announced they will be studying #3 - a 3rd shot / booster, as well as a booster with the new S. African mutant spike protein. Will be important studies.
I think many people think Covid will continue to mutate and be with us for a while. Leronlimab as immunomodulatory drug would theoretically be good for any respiratory virus causing hyperinflammatory response, so it could be helpful potentially for flu-related ARDS and others too, perhaps.
