I wouldn't rush to judgement with regard to a lack of efficacy in the first cohort.  We don't know how potent turning on the P53 will be.  The efficacy could be more than we know, even at low doses.  Assumption is the mother of all flock ups.  If a PR comes out in a day or two, the back fills will start after 5 hits imo.
